Hello JoomlaShine,

Can I set-up my website with JNS to allow visitors
to upload their own videos and add comments.

Does it offer seo friendly urls.

JSN Epic template (http://www.joomlashine.com/index.php?option=com_content&task=view&id=42&Itemid=38) represents only design’s part of your Joomla! CMS web site and do not effect to functionality of system. You can use JSN Epic template with one of extensions supporting video/audio functions.

Regarding to SEO urls, you can use the core Joomla! SEO parameter or 3rd party extensions. JSN Epic template support works fine in both cases.
